Static bioassays were conducted with pesticides like PP'-DDT, Dimethoate (Rogor) and Carbaryl (Sevin) to determine the median lethal concentrations (LC sub(50)) on an estuarine teleost Therapon jarbua (Forsk). The respiration rates of fishes exposed to pesticides, as well as those of controls were determined. Respiration abnormalities were noticed in treated fishes. The metabolic rates are generally higher in treated fishes than in the controls. The behaviour of fishes exposed to LC sub(25) (96h) concentrations of pesticides is discussed. Estuarine fishes appear to be more sensitive and susceptible to pesticides than fresh water fishes. The pesticides affect the locomotory and swimming behaviour of fishes. Loss in weight of fishes exposed to LC sub(50) (96 h) concentration of pesticides was also estimated. The present report gives a comprehensive account of the toxic nature of these pesticides to fishes.

The cardiomyocyte is a complex biological system where many mechanisms interact non-linearly to regulate the coupling between electrical excitation and mechanical contraction. For this reason, the development of mathematical models is fundamental in the field of cardiac electrophysiology, where the use of computational tools has become complementary to the classical experimentation. My doctoral research has been focusing on the development of such models for investigating the regulation of ventricular excitation-contraction coupling at the single cell level. In particular, the following researches are presented in this thesis: 1) Study of the unexpected deleterious effect of a Na channel blocker on a long QT syndrome type 3 patient. Experimental results were used to tune a Na current model that recapitulates the effect of the mutation and the treatment, in order to investigate how these influence the human action potential. Our research suggested that the analysis of the clinical phenotype is not sufficient for recommending drugs to patients carrying mutations with undefined electrophysiological properties. 2) Development of a model of L-type Ca channel inactivation in rabbit myocytes to faithfully reproduce the relative roles of voltage- and Ca-dependent inactivation. The model was applied to the analysis of Ca current inactivation kinetics during normal and abnormal repolarization, and predicts arrhythmogenic activity when inhibiting Ca-dependent inactivation, which is the predominant mechanism in physiological conditions. 3) Analysis of the arrhythmogenic consequences of the crosstalk between β-adrenergic and Ca-calmodulin dependent protein kinase signaling pathways. The descriptions of the two regulatory mechanisms, both enhanced in heart failure, were integrated into a novel murine action potential model to investigate how they concur to the development of cardiac arrhythmias. These studies show how mathematical modeling is suitable to provide new insights into the mechanisms underlying cardiac excitation-contraction coupling and arrhythmogenesis.

Chemokines are small, secreted proteins that orchestrate the migration of cells, which are involved in immune defence, immune surveillance and haematopoiesis. However, chemokines are also implicated in the pathology of various inflammatory diseases, cancers and HIV. The chemokine system is considerably large and has a redundancy in the repertoire of its inflammatory mediators. Therefore, strict regulation of chemokine activity is crucial. Chemokines are the substrate for various proteases including the serine protease CD26/dipeptidyl-peptidase IV and matrix metalloproteinases. Regulation by proteolytic cleavage controls and fine-tunes chemokine function by either enhancing or reducing its chemotactic activity or receptor selectivity. Often chemokines and the proteases that regulate them are produced in the same microenvironment and expression of both may be simultaneously induced by a common stimulus enabling the rapid regulation of chemokine activity. The overall impact of cleaved chemokines in cellular responses is very complex. In this review, we will give an overview on chemokine modification and the respective chemokine modifying proteases. Furthermore, we will summarize the emerging literature describing the consequences in inflammation, haematopoiesis, cancer and HIV infection upon proteolytic chemokine processing.

There were three principle aims to this thesis. Firstly, the acquisition protocols of clinical blood flow apparatus were investigated in order to optimise them for both cross-sectional and longitudinal application. Secondly, the effects of physiological factors including age and systematic circulation on ocular blood flow were investigated. Finally, the ocular perfusion characteristics of patients diagnosed with ocular diseases considered to be of a vascular origin were investigated. The principle findings of this work are:- 1) Optimisation of clinical investigationsPhotodiode sensitivity of the scanning laser Doppler flowmeter should be kept within a range of 70-150 DC when acquiring images of the retina and optic nerve head in order to optimise the reproducibility of capillary blood flow measures. Account of the physiological spatial variation in retinal blood flow measures can be made using standard analysis protocols of the scanning laser Doppler flowmeter combined with a local search strategy. Measurements of pulsatile ocular blood flow using the ocular blood flow analyser are reproducible, however this reproducibility can be improved when consecutive intraocular pressure pulses are used to calculate pulsatile ocular blood flow. Spectral analysis of the intraocular pressure pulse-wave is viable and identifies the first four harmonic components of the waveform. 2) Physiological variation in ocular perfusionAge results in a significant reduction in perfusion of the retinal microcirculation, which is not evident in larger vessel beds such as the choroid. Despite known asymmetry in the systemic vasculature, no evidence of interocular asymmetry in ocular perfusion is apparent. 3) Pathological variation in ocular perfusionIn primary open angle glaucoma, perfusion is reduced in the retinal microcirculation of patients classified as having early to moderate visual field defects. However, ocular pulsatility defects are masked when patients and subjects are matched for systemic variables (pulse rate and mean arterial pressure); differentiation is facilitated by the application of waveform analysis to the continuos intraocular pressure curve even in the early stages of disease. Diabetic patients with adequate glycaemic control, exhibit maintenance of macular blood flow, macular topography and visual function following phacoemulsification.

Deaths from microcystin toxication have widely been attributed to hypovolemic shock due to hepatic interstitial hemorrhage, while some recent studies suggest that cardiogenic complication is also involved. So far, information on cardiotoxic effects of MC has been rare and the underlying mechanism is still puzzling. The present study examined toxic effects of microcystins on heart muscle of rats intravenously injected with extracted MC at two doses, 0.16LD(50) (14 mu g MC-LReq kg(-1) body weight) and 1LD(50) (87 mu g MC-LReq kg(-1) body weight). In the dead rats, both TTC staining and maximum elevations of troponin I levels confirmed myocardial infarction after MC exposure, besides a serious interstitial hemorrhage in liver. In the 1LD(50) dose group, the coincident falls in heart rate and blood pressure were related to mitochondria dysfunction in heart, while increases in creatine kinase and troponin I levels indicated cardiac cell injury. The corresponding pathological alterations were mainly characterized as loss of adherence between cardiac myocytes and swollen or ruptured mitochondria at the ultrastructural level. MC administration at a dose of 1LD(50) not only enhanced activities and up-regulated mRNA transcription levels of antioxidant enzymes, but also increased GSH content. At both doses, level of lipid peroxides increased obviously, suggesting serious oxidative stress in mitochondria. Simultaneously. complex I and III were significantly inhibited, indicating blocks in electron flow along the mitochondrial respiratory chain in heart. In conclusion, the findings of this study implicate a role for MC-induced cardiotoxicity as a potential factor that should be considered when evaluating the mechanisms of death associated with microcystin intoxication in Brazil. The vertebral column and its units, the vertebrae, are fundamental features, characteristic of all vertebrates. Developmental segregation of the vertebral bodies as articulated units is an intrinsic requirement to guarantee the proper function of the spine. Whenever these units become fused either during development or postsegmentation, movement is affected in a more or less severe manner, depending on the number of vertebrae affected. Nevertheless, fusion may occur as part of regular development and as a physiological requirement, like in the tetrapod sacrum or in fish posterior vertebrae forming the urostyle. In order to meet the main objective of this PhD project, which aimed to better understand the molecular and cellular events underlying vertebral fusion under physiological and pathological conditions, a detailed characterization of the vertebral fusion occurring in zebrafish caudal fin region was conducted. This showed that fusion in the caudal fin region comprised 5 vertebral bodies, from which, only fusion between [PU1++U1] and ural2 [U2+] was still traceable during development. This involved bone deposition around the notochord sheath while fusion within the remaining vertebral bodies occur at the level of the notochord sheath, as during the early establishment of the vertebral bodies. A comparison approach between the caudal fin vertebrae and the remaining vertebral column showed conserved features such as the presence of mineralization related proteins as Osteocalcin were identified throughout the vertebral column, independently on the mineralization patterns. This unexpected presence of Osteocalcin in notochord sheath, here identified as Oc1, suggested that this gene, opposing to Oc2, generally associated with bone formation and mature osteoblast activity, is potentially associated with early mineralization events including chordacentrum formation. Nevertheless, major differences between caudal fin region and anterior vertebral bodies considering arch histology and mineralization patterns, led us to use RA as an inductive factor for vertebral fusion, allowing a direct comparison of equivalent structures under normal and fusion events. This fusion phenotype was associated with notochord sheath ectopic mineralization instead of ectopic perichordal bone formation related with increased osteoblast activity, as suggested in previous reports. Additionally, alterations in ECM content, cell adhesion and blood coagulation were discussed as potentially related with the fusion phenotype. Finally, Matrix gla protein, upregulated upon RA treatment and shown to be associated with chordacentrum mineralization sites in regular development, was further described considering its potential function in vertebral formation and pathological fusion. Therefore with this work we propose zebrafish caudal fin vertebral fusion as a potential model to study both congenital and postsegmentation fusion and we present candidate factors and genes that may be further explored in order to clarify whether we can prevent vertebral fusion.

The division of labor is a central theme in the study of social insects. In bees and wasps, this activity is regulated by age polyethism. Important physiological and morphological changes have been widely studied in the polyethism of honeybee workers. In contrast, this is a relatively unexplored subject in social vespids. Our goal was to determine if there are detectable morphological changes in the body of the Epiponini wasp Polybia paulista Von Ihering or in certain glands in relation to age polyethism. We observed changes in the body weight, the salivary gland, and the mandibular gland that were associated with age, and our results suggest that social relationships and task performance are important to these changes. This contrasts with observations in Polistes and is different from the Apis mellifera Linnaeus age polyethism model. © 2013 Sociedade Entomológica do Brasil.

Introduction: Life expectancy is increasing and becoming a characteristic phenomenon of developed countries and, increasingly, of developing countries, such as Brazil. The aging process causes changes of some physiological functions such as loss of smell, taste, loss of appetite, among other things that end up changing the food intake of these individuals. Objectives: This study aimed to assess food consumption of the young and long-lived elderly in a city in southern Brazil. Methods: A cross-sectional survey conducted through home visits in Palmeira das Missões - RS, Brazil. The sociodemographic, anthropometrical and dietary data were collected through questionnaires and 24-hour recall. The adequacy of nutrients was assessed according to the Dietary Reference Intakes. Data were analyzed using SPSS 18.0 software. Results: The study included 424 older adults, 84,4% (n = 358) aged less than 80 years old and 15,6% (n = 66) older than 80. The intake of energy and protein was insufficient for both young elderly and the oldest. The consumption of vitamins and minerals has been insufficient in all seniors except for iron, which presented an excessive intake. There was a statistically significant difference between the elderly and oldest only for the consumption of lipids and vitamin B12. Conclusion: The majority of studies with elderly corroborate the results found in this article. An inadequate intake of nutrients can develop nutritional deficiencies, and consequently it can result in physiological and pathological changes which would compromise the functional capacity of the elderly. Energy consumption was insufficient and macronutrients were inadequate, both for the young elderly as for the oldest. Additionally, the consumption of vitamins and minerals was insufficient to everyone except the iron, which presented excessive intake for young and oldest elderly.

干扰素(IFNs)是最早发现的具有广泛用途的一类细胞因子,IFN-α通过JAK/STAT信号途径调控机体一系列生理和病理反应。至今尚未发现类干扰素的小分子。我们前期研究发现天然产物毛蕊异黄酮可激活干扰素诱导的JAK/STAT信号途径。为发现类干扰素小分子、获得小分子探针,本课题拟建立成熟的JAK/STAT信号途径的筛选模型,合成毛蕊异黄酮及其类似物,研究这些化合物的构效关系,进而尝试通过共价键标记生物素或香豆素来直接研究它们与相关受体的作用。 从异香草醛出发经7步合成反应得到了毛蕊异黄酮。采用平行合成策略得到异黄酮类化合物;采用分支式合成策略,以取代苯乙酸作为合成砌块,获得具有与异黄酮类似结构的香豆素、3-芳基喹诺酮。与分离得到的黄酮类化合物,构建了一个包括异黄酮、黄酮、香豆素、3-芳基喹诺酮在内的化合物库。 建立了包含IFN-α刺激反应元件 (ISRE)的荧光素酶报告基因体系,通过筛选化合物库中的化合物,发现异黄酮骨架为激活JAK/STAT信号途径必须结构、毛蕊异黄酮7-位酚羟基被取代后活性丧失。根据以上结果,对毛蕊异黄酮3′-位标记物的合成进行了初步尝试。 发现山茱萸科植物青荚叶(Helwingia japonica (Thunb.) Dietr.)有抑制蛋白酪氨酸磷酸酯酶1B(PTP1B)的活性。从其地上部分95%乙醇提取物的乙酸乙酯部分分离得到5个化合物,应用波谱方法及与已知品对照的手段鉴定它们为p-menth-2-en-1β, 4β, 8-triol (Z-1)、blumenol A (Z-2)、2′,3′,4′,5′,6′-五羟基查尔酮(Z-3)、洋芹素7-O-β-D-吡喃葡萄糖苷(Z-4)、木犀草素7-O-β-D-吡喃葡萄糖苷(Z-5). Interferons (IFNs) are one kind of cytokines with broad functions. IFN-α mediates series physiological and pathological changes of human body via JAK/STAT pathway. Untill now, no IFNs-like small molecules are discovered. In our preliminary experiment, the natural product calycosin has been observed to activate JAK/STAT pathway. Therefore, we establish a luciferase reporter gene system and synthesize calycosin and its analogues to reveal their structure-activity relationship (SAR). Besides, in order to prove that calycosin activates JAK/STAT pathway through IFN receptor, we attempted to tag it with biotin or coumarin by covalent bonding. Calycosin was synthesized from isovanillin via seven steps. Other isoflavones were obtained by parallel synthesis; coumarins and quinolones were prepared through divergent synthesis, using substituted phenylacetic acids as building blocks. Combing with natural flavones, a small molecule library was established. A luciferase reporter gene system, consisting of 5 copies of the ISRE (interferon-stimulated response element), was used for screening of small molecules from that library. We found that the core-structure of isoflavone was necessary, and if the 7-OH is substituted, the activity slumps. According to our observation, we tried to tag biotin or coumarin at 3′-OH of calycosin. The 95% ethanol extract of the aerial parts of Helwingia japonica (Thunb.) Dietr. showed protein tyrosine phosphatase 1B (PTP1B) inhibitory activity. Five compounds were isolated. On the basis of spectral data or by comparison with authentic samples, they were identified as p-menth-2-en-1β,4β,8-triol (1), blumenol A (2), 2′,3′,4′,5′,6′-pentahydroxychalcone (3), apigenin 7-O-β-D-glucopyranoside (4), and luteolin 7-O-β-D-glucopyranoside (5).

The practice of physical exercise and the adopting of a healthy lifestyle are some of the ways to minimize the physiological and pathological changes associated with aging. Therefore, it is interesting to search for new technologies that encourage the elderly to practice physical activities, such as Exergames. In this study, an integrative review was carried out through electronic databases, journals and scientific journals searching for articles and publications that relate the use of exergames by retirees and testing the viability of these technologies towards this target public. A total of twenty-seven articles made part of the review. After analyzing the results, it's clear that exergames are one of the pertinent options to motivate this public to adhere in a healthier lifestyle, being able to improve the physical, cognitive abilities and improve emotional states of its practitioners, can vary the intensity of exercise in accordance with the personal limitations of each one. But certain care and adjustments need to be made before starting to practice them
